Semax is a synthetic heptapeptide (MEHFPGP) derived from the ACTH(4-7) sequence, developed in Russia at the Institute of Molecular Genetics. It is a well-characterised neuropeptide analogue with a proline-glycine-proline (PGP) C-terminal extension that confers resistance to enzymatic degradation, significantly extending its biological half-life compared to the parent ACTH fragment. Semax has been extensively studied in preclinical research for its neurotrophic and neuroprotective properties, and is one of the most thoroughly investigated neuropeptides in the Russian scientific literature.
Semax has been studied in preclinical models for its effects on brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) expression in the hippocampus and frontal cortex, supporting neuronal survival and synaptic plasticity. It also modulates dopaminergic and serotonergic systems, and has been shown to influence melanocortin receptor signalling via its ACTH-derived sequence. Additionally, Semax has been investigated for its effects on pro-inflammatory cytokine expression and oxidative stress markers in neuronal tissue.
