Semax

Semax is a synthetic heptapeptide (MEHFPGP) derived from the ACTH(4-7) sequence, developed in Russia at the Institute of Molecular Genetics. It is a well-characterised neuropeptide analogue with a proline-glycine-proline (PGP) C-terminal extension that confers resistance to enzymatic degradation, significantly extending its biological half-life compared to the parent ACTH fragment. Semax has been extensively studied in preclinical and clinical research for its neurotrophic, neuroprotective, and cognitive-enhancing properties, and is one of the most thoroughly investigated nootropic peptides in the Russian scientific literature.

Mechanism of Action

Semax exerts its primary effects through upregulation of brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) expression in the hippocampus and frontal cortex, supporting neuronal survival, synaptic plasticity, and long-term potentiation. It also modulates the dopaminergic and serotonergic systems, and has been shown to influence melanocortin receptor signalling via its ACTH-derived sequence. Additionally, Semax demonstrates anti-inflammatory properties through inhibition of pro-inflammatory cytokine expression and attenuation of oxidative stress in neuronal tissue.
