Selank

Selank is a synthetic heptapeptide (TKPRPGP) developed at the Institute of Molecular Genetics of the Russian Academy of Sciences. It is a structural analogue of the endogenous immunomodulatory tetrapeptide tuftsin (TKPR), extended with a proline-glycine-proline (PGP) C-terminal sequence to enhance metabolic stability and prolong biological activity. Selank has been extensively studied in preclinical and clinical research for its anxiolytic, nootropic, and immunomodulatory properties, and is one of the most thoroughly characterised peptide anxiolytics in the Russian scientific literature.

Mechanism of Action

Selank exerts its anxiolytic effects primarily through modulation of GABAergic neurotransmission, increasing the affinity of GABA-A receptors for GABA without direct benzodiazepine-site binding — producing anxiolysis without sedation or dependence. It also inhibits the enzymatic degradation of endogenous enkephalins, prolonging their activity at opioid receptors and contributing to mood stabilisation. Additionally, Selank upregulates BDNF expression in the hippocampus, modulates IL-6 and other cytokine levels, and influences serotonergic and dopaminergic neurotransmission, supporting its investigation across anxiety, cognition, and neuroimmune research.
