GHK-Cu (Glycyl-L-histidyl-L-lysine copper complex) is a research-grade naturally occurring copper-binding tripeptide found in human plasma, saliva, and urine, developed for preclinical investigation of wound healing, tissue remodelling, anti-inflammatory signalling, and gene expression regulation. First isolated by Loren Pickart in 1973, GHK-Cu has been extensively studied for its ability to stimulate collagen and glycosaminoglycan synthesis, modulate metalloproteinase activity, and regulate a broad array of genes associated with tissue repair and antioxidant defence.
Mechanism of Action
GHK-Cu acts through multiple mechanisms: it chelates copper(II) ions and delivers them to cells, facilitating copper-dependent enzymatic activity including lysyl oxidase-mediated collagen crosslinking. It modulates the expression of over 4,000 human genes, upregulating pathways involved in tissue repair, anti-inflammatory signalling, and antioxidant defence, while downregulating genes associated with inflammation and cancer progression. GHK-Cu also stimulates the production of collagen, elastin, and glycosaminoglycans, and activates matrix metalloproteinases (MMPs) to facilitate the removal of damaged extracellular matrix components.
