Vasoactive Intestinal Peptide (VIP) is a 28-amino acid neuropeptide belonging to the secretin/glucagon superfamily. First isolated from porcine intestinal tissue, VIP is widely distributed throughout the central and peripheral nervous systems and exerts pleiotropic effects on smooth muscle relaxation, immune modulation, and neurotransmission. Its broad receptor distribution — primarily via VPAC1 and VPAC2 receptors — makes it a subject of significant interest in preclinical research across multiple physiological systems.
VIP binds with high affinity to VPAC1 and VPAC2 G-protein-coupled receptors, activating adenylyl cyclase and elevating intracellular cAMP. This cascade mediates smooth muscle relaxation, vasodilation, and immunomodulatory effects including suppression of pro-inflammatory cytokine release. VIP has also been shown to promote neuronal survival and modulate circadian rhythm signalling via the suprachiasmatic nucleus.
