Matrixyl, chemically designated as Palmitoyl Pentapeptide-4 (Pal-Lys-Thr-Thr-Lys-Ser), is a synthetic lipopeptide consisting of a pentapeptide sequence derived from the C-terminal domain of type I procollagen, conjugated to a palmitoyl fatty acid chain to enhance skin penetration and stability. It was developed by Sederma as a cosmetic active ingredient and has been extensively studied in dermatological and cell biology research for its ability to stimulate extracellular matrix (ECM) protein synthesis in fibroblasts. Mechanism of Action

Matrixyl acts as a matrikine — a peptide fragment derived from ECM proteins that signals to cells to regulate matrix remodelling. The pentapeptide sequence (KTTKS) is a fragment of the type I procollagen α-chain that binds to fibroblast surface receptors, activating TGF-β-like signalling pathways and stimulating the synthesis of collagen I, collagen III, fibronectin, and hyaluronic acid. The palmitoyl conjugation enhances lipophilicity, facilitating membrane interaction and cellular uptake. Matrixyl also modulates matrix metalloproteinase (MMP) activity, influencing ECM turnover and tissue remodelling dynamics, making it a useful tool for studying fibroblast biology, wound healing, and dermal matrix homeostasis.
