Kisspeptin-10 is a synthetic decapeptide corresponding to the C-terminal 10 amino acids of the kisspeptin family of neuropeptides, which are encoded by the KISS1 gene. Kisspeptins are endogenous ligands of the G-protein-coupled receptor GPR54 (also known as KISS1R), expressed predominantly in the hypothalamus, pituitary, and gonads. Kisspeptin-10 represents the minimal bioactive fragment of the kisspeptin family and is the most potent and widely studied isoform in reproductive neuroendocrinology research. It is a critical regulator of the hypothalamic-pituitary-gonadal (HPG) axis and has been extensively investigated in preclinical and clinical research for its roles in GnRH pulse generation, puberty onset, and fertility regulation.

Mechanism of Action

Kisspeptin-10 acts as a potent agonist of GPR54 (KISS1R), a Gq/11-coupled receptor expressed on GnRH neurons in the hypothalamic arcuate nucleus (ARC) and anteroventral periventricular nucleus (AVPV). GPR54 activation stimulates phospholipase C (PLC), generating IP3 and DAG, which increase intracellular calcium and activate PKC, leading to membrane depolarisation and GnRH pulse release. GnRH then stimulates pituitary gonadotroph cells to secrete LH and FSH, which drive gonadal steroidogenesis and gametogenesis. Kisspeptin neurons in the ARC also integrate metabolic signals (leptin, insulin, ghrelin) and sex steroid feedback, making the kisspeptin-GPR54 axis a key regulatory node for reproductive function, energy balance, and pubertal timing.
