SS-31, also known as Elamipretide or MTP-131, is a synthetic tetrapeptide (D-Arg-2’,6’-Dmt-Lys-Phe-NH2) belonging to the Szeto-Schiller (SS) peptide family. It is specifically designed to target and concentrate within the inner mitochondrial membrane, where it interacts with cardiolipin — a phospholipid critical for mitochondrial cristae architecture and electron transport chain function. SS-31 has emerged as a leading research tool in mitochondrial biology, with a substantial body of preclinical literature supporting its investigation across ageing, ischaemia-reperfusion injury, and metabolic disease models.
SS-31 selectively partitions into the inner mitochondrial membrane due to its alternating aromatic and cationic residues, achieving mitochondrial concentrations several hundred-fold above cytoplasmic levels. It binds directly to cardiolipin, stabilising cristae morphology and preserving the structural integrity of electron transport chain supercomplexes (respirasomes). This interaction reduces mitochondrial reactive oxygen species (ROS) production, restores ATP synthesis efficiency, and inhibits cytochrome c peroxidase activity — collectively attenuating mitochondrial dysfunction and apoptotic signalling.
