Snap-9 (also referred to as Acetyl Glutamyl Heptapeptide-3) is a synthetic nonapeptide developed as a cosmetic research compound targeting the neuromuscular junction. It is a structural analogue of the SNAP-25 protein — a key component of the SNARE complex responsible for acetylcholine vesicle docking and fusion at the neuromuscular synapse. Snap-9 has been investigated in in vitro models for its capacity to modulate neurotransmitter release and attenuate muscle contraction signalling, making it a subject of interest in cosmetic peptide and dermatological research.
Snap-9 competitively inhibits the formation of the SNARE complex by mimicking the SNAP-25 binding domain, thereby interfering with the docking and fusion of acetylcholine-containing synaptic vesicles at the neuromuscular junction. This reduces acetylcholine release and attenuates the downstream signal cascade that triggers muscle fibre contraction. In vitro studies have demonstrated dose-dependent inhibition of muscle contraction in cell-based models, supporting its investigation as a topical neuromodulatory peptide in cosmetic research contexts.
