PNC-27

PNC-27 is a synthetic anticancer peptide derived from the MDM-2-binding domain of p53, the tumour suppressor protein. It was developed as a research tool to investigate selective cancer cell membrane disruption, exploiting the differential expression of HDM-2 (the human homologue of MDM-2) on the plasma membranes of cancer cells versus normal cells. Mechanism of Action

PNC-27 contains a p53 residue 12-26 sequence fused to a membrane-penetrating leader peptide. It is hypothesised to bind to HDM-2 expressed on the extracellular surface of cancer cell membranes — a phenomenon not observed in normal cells — triggering pore formation and direct membrane disruption leading to necrotic cell death.
