Mechanism of Action
Gonadorelin binds to GnRH receptors (GnRHR) on pituitary gonadotroph cells, activating Gq/11-coupled phospholipase C signalling and triggering intracellular calcium release and protein kinase C activation. This leads to the synthesis and pulsatile secretion of LH and FSH, which in turn stimulate gonadal steroidogenesis and gametogenesis. The pulsatile nature of GnRH signalling is critical: continuous exposure leads to receptor downregulation and gonadotropin suppression, while pulsatile administration maintains or stimulates the HPG axis — a distinction of significant research relevance.
