Peptide Reconstitution Guide: A Step-by-Step Laboratory Protocol

Introduction

Lyophilised (freeze-dried) peptides must be reconstituted before use in laboratory assays. Correct reconstitution technique is critical to preserving peptide integrity, ensuring accurate concentration, and maintaining stability during storage.

Materials Required

  • Lyophilised peptide vial
  • Bacteriostatic Water (BW) — preferred for most peptides
  • 0.6% Acetic Acid Solution — for poorly soluble peptides (e.g. IGF-1 LR3, HGH Fragment)
  • Sterile syringes and needles
  • Alcohol swabs
  • Refrigerator (2–8°C) or freezer (−20°C) for storage

Choosing the Right Solvent

  • Bacteriostatic Water — suitable for BPC-157, TB-500, Semaglutide, Tirzepatide, CJC-1295, Ipamorelin and most GH secretagogues.
  • 0.6% Acetic Acid — recommended for IGF-1 LR3, IGF-DES, and HGH Fragment 176-191.
  • Sterile Water — short-term use only. Significantly reduces shelf life.

Reconstitution Protocol

  1. Allow the vial to reach room temperature — 10–15 minutes before opening.
  2. Prepare your solvent — draw the required volume into a sterile syringe.
  3. Clean the vial septum — wipe with an alcohol swab, air dry 30 seconds.
  4. Inject solvent slowly down the side of the vial — never directly onto the lyophilised cake.
  5. Do not shake — gently swirl until fully dissolved.
  6. Inspect the solution — should be clear and colourless. Discard if cloudy or particulate.

Calculating Concentration

Concentration (mg/mL) = Peptide mass (mg) ÷ Volume of solvent added (mL)

Example: A 5 mg vial + 2 mL bacteriostatic water = 2.5 mg/mL.

Storage After Reconstitution

  • Bacteriostatic water: 2–8°C for up to 28 days.
  • Longer storage: aliquot and freeze at −20°C. Avoid repeated freeze-thaw cycles.
  • Lyophilised (unreconstituted): stable 24+ months at −20°C, kept dry and away from light.
