FOXO4-DRI is a research-grade D-amino acid retro-inverso peptide designed for preclinical investigation of cellular senescence, apoptosis, and tissue rejuvenation. Developed by Peter de Keizer and colleagues, FOXO4-DRI is a modified peptide that disrupts the interaction between the FOXO4 transcription factor and p53 in senescent cells, selectively triggering apoptosis in cells that have entered a state of permanent cell cycle arrest. It represents a significant tool compound in the emerging field of senolytic research.
In senescent cells, FOXO4 forms a complex with p53 that sequesters p53 in the nucleus and prevents apoptosis, allowing senescent cells to persist and secrete pro-inflammatory factors (the senescence-associated secretory phenotype, SASP). FOXO4-DRI competitively disrupts this FOXO4–p53 interaction, freeing p53 to translocate to the mitochondria and initiate the intrinsic apoptotic cascade selectively in senescent cells. Non-senescent cells, which do not rely on this FOXO4–p53 interaction for survival, are largely unaffected, conferring a degree of selectivity that makes FOXO4-DRI a valuable senolytic research tool.
