Adipotide (also known as FTTP — Fatty acid-Targeted Proapoptotic Peptide — and PROHIBIT) is a proapoptotic research peptide designed to selectively target and destroy the vasculature supplying white adipose tissue. It is a chimeric peptide consisting of a targeting domain (CKGGRAKDC) that binds to prohibitin on the surface of adipose blood vessel endothelial cells, linked to a proapoptotic domain (D(KLAKLAK)₂) that disrupts mitochondrial membranes upon internalisation.
Mechanism of Action
The targeting sequence CKGGRAKDC homes specifically to prohibitin expressed on the luminal surface of blood vessels within white adipose tissue. Upon binding, the proapoptotic KLAKLAK₂ domain is internalised and disrupts mitochondrial membranes, triggering apoptosis in the targeted endothelial cells. This leads to localised vascular regression and subsequent apoptosis of the dependent adipocytes, resulting in fat mass reduction in research models without systemic toxicity at studied doses.
